Humoral and cellular immune responses in critically ill influenza A/H1N1‐infected patients

Abstract There is limited knowledge of influenza‐specific immune responses and their kinetics in critically ill patients. We investigated humoral cellular after critical influenza A/H1N1 infection hypothesized that dysfunctionality or absence could contribute to more severe illness. followed 12 patients hospitalized with infection; the majority admitted intensive care unit (ICU). Blood samples were collected at days 10 19 5 months. Antibody surface glycoproteins haemagglutinin (HA) neuraminidase (NA) A/H1N1pdm09 quantified by haemagglutination inhibition (HAI), microneutralization (MN), Enzyme‐linked immunosorbent assay (ELISA) lectin (ELLA). Influenza‐specific antibody levels avidity measured separately for head stalk domains H1. Cytokine secreting CD4 + CD8 T cell conserved epitopes (M1, NP PB1) analysed FluoroSpot. Overall, retained a high level functional HA‐ NA‐specific antibodies over study period. During acute phase (up 3 weeks from symptom onset), specific H1 increased earlier present higher amount compared head‐specific antibodies. The non‐neutralizing HA‐specific response full‐length showed significant decline convalescent phase. Despite total IgG concentrations, protein remained low all time points. Similarly, continuously levels. In conclusion, our found characterized response.
